I was looking for a new area to DM in and was curious on Erlkazar. As I'm doing searches on everything I can find out about the place, I come upon the fact that it's now ruled by vampires.

While I know how and why, what I don't know is when? Is that event a 4E storyline, meaning it takes place 100 years later, or is it something that happenened pre-Spellplague? Mention of King Korox has me assuming pre-Spellplague but not sure when. Is it 1374 or 1375 or later?

Not really sure, thought it's left vague on purpose so you can fill in whatever story your like to make it work. Personally, I like the idea of elements of the Chaos Curse (Cleric Quintet series) to have affected a portion of the people there.

I don't think it's directly related to the Spellplague

well, we know that Erlkazar was plagued by vampires starting in 1348 from Lands of Intrigue:

What happened after 1358 is anyone's guess....

We also know that a very powerful vampire, over 800 years, had a lair there from LoI:

Though she would not likely be involved - at least not directly - with ruling Erlkazar. The Rune prefers to manipulate the strings from afar. Even putting her own vampires into ruling would be too close to the Twisted Rune.

I agree... Though I can see her "encouraging" other vampires to come to the area, to keep anything she does from being noticed.

The 4th edition lore on the subject seems a bit conflicting. On the one hand, the master vampire in charge of it all is one Saestra Karanok who was driven from her home when her father discovered her nature.

However the Karanoks are from Luthcheq and that city whilst now the only "Point of Light in an otherwise monster infested Chessenta, it is also in an area perfect for vampire habitation. When the portions of the Underdark collapsed (one can only assume with the Spellplague) the Smoking Mountains nearby fired up again and permanently choke the sky with clouds so much so that a vampiric dragon is able to operate in the daytime.

Now either she is a recent (post Spellplague) exile and didnt settle in Chessenta because of the vampire dragon (who probably doesnt want any competition from a vampire that isnt under his control).

Or she fled Luthcheq and Chessenta pre spellplague and travelled around a lot before settling in Erlkazar. Now the 4th edition lore would seem to support this as it mentions she travelled for years (non specific i know but with other paragraphs supporting this theory), it also mentions that she has generations (which would imply she has been there a considerable amount of time) of bandits serving her, many are vampires.
Since i havent found any lore in 3.5 books it is possible that she was discovered and fled sometime between 1375 and 1385 (or whenever the spellplague begins), which gives plenty of time (a decade) for a child to come of age and become a vampire and be discovered and exiled all before her homeland becomes the perfect place for a vampire to live.

After travelling for years the spell plague hits and she uses the confusion and inevitable anarchy that follows to set up home in unlucky Erlkazar.

Anyway thats just my two pence on the possibilities

Saestra Karanok was a vampire back in 1357.

Out of "Old Empires" with the timeline showing 1357 as the present there is a blurb on her:

By the time she exited, she was a vampiress. She has successfully concealed this fact from her family even when she manipulated them into killing her vampire master. She has two personalities#151;one is sweet and totally innocent and loving, and the other is a savage killer. Either personality can come out at a moment#146;s notice, depending on her hunger.


Also in Dragon 236 she appears to be in her 20s (the age she was made a vampire):

Appearance
Standing at 5’9” tall, Saestra Karanok is a sultry brunette whose light violet eyes sparkle with glee whenever she is happy — or whenever she is about to feed on the living. She appears to be in her mid-20s.
As the daughter of the ruler of Luthcheq, she dresses in impeccably tailored color coordinated dresses and gowns which are usually black or purple and liberally decorated with gems, lace, and ribbons. She wears an ornamental dagger at her side.

I agree... Though I can see her "encouraging" other vampires to come to the area, to keep anything she does from being noticed.

That is, essentially, what Shyressa's attempting to accomplish as part of her overall plot in the Master of Chains novel.
