Why not?
Note: This primarily focuses on evil-aligned giants, namely the following
- Hill Giants
- Frost Giants
- Fire Giants
- Ogres
- Ogre Mages
- Trolls
- Ettins
- Verbeeg

So here's the party I was thinking of.
1) Barbarian with levels in the Giant-Killer prestige class (Silver Marches).

2) Ranger with levels in Foe Hunter (Masters of the Wild, obviously keyed to Giants) and/or Giant-Killer, with Giants as top Favored Enemy.

3) Cleric with the Protection Domain, possibly Strength as well. Utilizes buffing and healing spells.

4) Rogue, possibly a subrace of Gnome with levels in Gnome Giant-Killer (Complete Warrior), maybe even with levels in Blade Bravo (Races of Stone), focusing on landing Sneak Attacks to whittle a giant down.

5) Sorcerer or Wizard with spells to exploit elemental weaknesses of certain giants (ex: Fire for Frost Giants or Trolls, cold for Fire Giants, etc.) or to otherwise weaken them significantly as well as to buff their parties members (and any allies or cohorts they have).

Otherwise, you could throw in anything else you want, whether it be a Fighter (likely a Dwarf Fighter with levels in Dwarven Defender), Paladin, Bard, etc.. You could possibly also want a Druid instead of the Cleric but that could hamper buffing.
